In this article, we will cover all the aspects of Krazati to make you ready in and out for the treatment. If you are looking for Krazati tablets in India, knowing more about the treatment can help you better understand what to expect.

Science behind Krazati (Adagrasib)

When a mutation occurs in KRAS, a normal human gene, it is called KRAS G12C. This mutation makes cells grow and divide at a fast rate leading to the cancerous tumor. Krazati works by directly blocking the KRAS G12 C protein. The direct targeting action of Adagrasib in Krazati on the mutated KRAS G12 C genes slows and eventually stops tumor progression.

Eligible candidates for therapy

The eligible candidates for Krazati therapy according to Food and Drug Administration (FDA) are listed below:

  1. Non-small cell lung cancer (NSCLC)

As a single agent, for the treatment of adult patients with KRAS G12Cmutated locally advanced or metastatic NSCLC, as determined by an FDA-approved test, who have received at least one prior systemic therapy.

  1. Colorectal cancer (CRC)

In combination with cetuximab, for the treatment of adult patients with KRAS G12C-mutated locally advanced or metastatic CRC, as determined by an FDA-approved test, who have received prior treatment with fluoropyrimidine-, oxaliplatin-, and irinotecan-based chemotherapy

Dosage instructions

  • The recommended dosage of Krazati as a single agent or in combination with cetuximab is 600 mg orally twice daily until disease progression or unacceptable toxicity.
  • Maintain the same timing everyday to take the Krazati tablet every day with or without food.
  • Swallow tablets whole.
  • Do not chew, crush or split tablets.
  • Do not take extra tablets in case you vomit soon after taking it. Resume dosing at the next scheduled time.
  • If a dose is inadvertently missed, it should be skipped if greater than 4 hours have elapsed from the expected dosing time. Resume dosing at the next scheduled time.

Risks

  1. General:

Single agent use in NCSLC: The most common adverse reactions were nausea, diarrhea, vomiting, fatigue, musculoskeletal pain, hepatotoxicity, renal impairment, edema, dyspnea, and decreased appetite.
In combination with cetuximab in CRC: The most common adverse reactions were rash, nausea, diarrhea, vomiting, fatigue, musculoskeletal pain, hepatotoxicity, headache, dry skin, abdominal pain, decreased appetite, edema, anemia, and cough.

  1. Localised:

    • Gastrointestinal issues: Krazati can cause gastrointestinal bleeding, gastrointestinal obstruction in colitis, ileus and stenosis. Other symptoms include nausea, vomiting and diarrhoea. Monitor and manage patients using supportive care, including antidiarrheals, antiemetics, or fluid replacement, as indicated.
    • QT interval prolongation: Therapy can cause QTc interval prolongation, which can increase the risk for ventricular tachyarrhythmias (e.g., torsades de pointes) or sudden death. Monitor ECGs and electrolytes, particularly potassium and magnesium, before treatment during concomitant use, and as clinically indicated in patients with congestive heart failure, bradyarrhythmias, electrolyte abnormalities, and in patients who are unable to avoid concomitant medications that are known to prolong the QT interval.
    • Hepatotoxicity (liver toxicity): Patients taking Krazati can cause hepatotoxicity that can lead to drug-induced injury and hepatitis. Monitor liver laboratory tests (AST, ALT, alkaline phosphatase and total bilirubin) prior to the start of therapy and monthly for 3 months or as clinically indicated, with more frequent testing in patients who develop transaminase elevations.
    • Interstitial Lung Disease / Pneumonitis: This is usually fatal. It occurs in single-agent Krazati when compared to combination drugs. Monitor patients for dyspnoea (shortness of breathing), cough, fever and other worsening symptoms that are indicative of Interstitial Lung Disease / Pneumonitis

Understanding about the drug beforehand reduces anxiety and makes you stable throughout the therapy as you are aware of the expected risks. Get more updates on Krazati at 24/7qualityMeds.
