DOSAGE:

In the Phase I study published in the Journal of Hematology & Oncology, orally administered olverembatinib was evaluated at escalating alternate-day (QOD) doses ranging from 1 mg to 60 mg in 28-day treatment cycles.

ADVERSE EFFECTS:

Serious adverse events included thrombocytopenia, anemia, pneumonia, fever, and atrial fibrillation. 

Frequent nonhematologic treatment-related adverse events were changes in skin pigmentation and hypertriglyceridemia. Alterations in cutaneous pigmentation manifested as lentiginous nevi that did not require intervention or treatment discontinuation. The pathological mechanism has yet to be elucidated.

Potentially treatment-related cardiovascular events associated with olverembatinib included hypertension, pericardial effusion, ventricular extrasystoles, atrial fibrillation or supraventricular extrasystoles, retinal vein occlusion , and palpitations. Treatment-related angina, atrial tachycardia, cardiomegaly, arrhythmia, cerebral ischemia, and/or cerebral infarction were observed in <1% of patients each.

  4. Lipid and Metabolic Abnormalities: It can induce hypertriglyceridemia and hyperuricemia. Monitoring of lipid profiles and uric acid levels is advised to prevent metabolic and gout-related complications.
  5. Skin Pigmentation: The most common non-hematologic side effect is skin hyperpigmentation, which generally presents as harmless, tiny dark moles.
  6. Proteinuria: Mild to moderate proteinuria is frequently observed. Kidney function should be monitored throughout treatment.
  7. Pregnancy and Breastfeeding: Olverembatinib is contraindicated during pregnancy and breastfeeding, as it can cause fetal harm. Patients of childbearing potential should use effective contraception.
