Vitrakvi is indicated for the treatment of adult and pediatric patients with solid tumors that have a neurotrophic tyrosine receptor kinase (NTRK) gene fusion, without a known acquired resistance mutation. These tumors must be either metastatic or where surgical resection is likely to result in severe morbidity, and there must be no satisfactory alternative treatments or the cancer must have progressed following treatment. This indication is tumor-agnostic, meaning Vitrakvi is approved based on the presence of the NTRK gene fusion rather than the location or type of the cancer, making it suitable for various tumor types such as lung, thyroid, colon, and pediatric cancers like infantile fibrosarcoma.

Strength:

The capsules are available in strengths of 25mg and 100mg. Oral solution consists of 20mg/ml of larotrectinib. 

Dosage:

  • Adults and Pediatric Patients with Body Surface Area (BSA) ≥1 m²: 100 mg orally twice daily, with or without food, until disease progression or unacceptable toxicity occurs.
  • Pediatric Patients with BSA <1 m²: 100 mg/m² orally twice daily, with a maximum dose of 100 mg per dose, until disease progression or unacceptable toxicity occurs.
