Indication:Mylotarg is a targeted cancer therapy used to treat certain types of acute myeloid leukemia (AML), a fast-growing cancer of the blood and bone marrow. It is classified as an antibody-drug conjugate, meaning it combines a monoclonal antibody that specifically binds to CD33 - a protein commonly found on AML cells - with a powerful chemotherapy drug.

Mylotarg is indicated for the treatment of newly diagnosed CD33-positive acute myeloid leukemia (AML) in adults, either in combination with standard chemotherapy or as a single agent for patients aged 75 or older, or those who have comorbidities that preclude intensive chemotherapy. It is also approved for the treatment of relapsed or refractory CD33-positive AML in adults and in pediatric patients aged 1 month and older.

Dosage: 

Newly-diagnosed, de novo AML (combination regimen):  

  • Induction: 3 mg/m2 (up to one 4.5 mg vial) on Days 1, 4, and 7 in combination with daunorubicin and cytarabine. 
  • Consolidation: 3 mg/m2 on Day 1 (up to one 4.5 mg vial) in combination with daunorubicin and cytarabine.  

Newly-diagnosed AML (single-agent regimen):  

  • Induction: 6 mg/m2 on Day 1 and 3 mg/m2 on Day 8.  
  • Continuation: For patients without evidence of disease progression following induction, up to 8 continuation courses of Mylotarg 2 mg/m2 on Day 1 every 4 weeks.  

Relapsed or refractory AML(single-agent regimen):

  • 3 mg/m2 on Days 1, 4, and 7. 

Side effects

  • Fever, infection, bleeding, nausea, vomiting, constipation, rash, headache, liver problems, mouth sores, inflammation of the gut, and low white blood cell count (with or without fever).

  • Do not freeze
  • Fatal hepatotoxicity may occur in the patients who are on mylotarg treatment. 
  • Infusion related reactions may occur which could be fatal sometimes. This could occur during or within 24hrs of mylotarg infusion. Therefore, premedication is required before infusing Mylotarg. 
  • Mylotarg is observed to be myelosuppressive, which may cause life threatening  hemorrhage due to prolonged thrombocytopenia.
