Indication:Zavesca is an oral medication used to treat certain rare genetic disorders, most notably mild to moderate type 1 Gaucher disease in adults who cannot receive enzyme replacement therapy. It belongs to a class of drugs known as substrate reduction therapies. Zavesca helps reduce the symptoms and complications associated with the disease, such as enlarged organs, bone pain, and fatigue. It is taken by mouth, typically three times a day, and requires regular monitoring for side effects and treatment response.

Zavesca is indicated for the treatment of adult patients with mild to moderate type 1 Gaucher disease for whom enzyme replacement therapy (ERT) is not an option, either due to hypersensitivity, poor venous access, or other medical reasons. Type 1 Gaucher disease is a rare genetic disorder caused by a deficiency of the enzyme glucocerebrosidase, leading to the buildup of fatty substances in certain organs, particularly the spleen, liver, and bone marrow. Zavesca works as a substrate reduction therapy by inhibiting the enzyme glucosylceramide synthase, which reduces the production of the fatty substrate that accumulates in patients with Gaucher disease. It is used as an oral alternative when standard ERT is unsuitable or unavailable.

Dosage: 

100 mg capsule administered orally three times a day at regular intervals. It may be necessary to reduce the dose to one 100 mg capsule once or twice a day in some patients due to adverse reactions, such as tremor or diarrhea. 

Patients with Renal Impairment needs dosage adjustment:

  • For mild renal impairment: 100 mg capsule twice per day
  • For moderate impairment:  one 100 mg capsule per day
  • Zavesca is not recommended in patients with severe renal impairment.  

Storage: 

  • Store the cartons and blister packs of Zavesca at 20°C to 25°C 
  • Keep out of reach of children.
  • Peripheral Neuropathy has been observed in patients and due to the same, there should be a continuous neurological monitoring of patients. 
  • Tremors usually began at initial treatment therapy, dose reduction or discontinuation of treatment to the patient taking Zavesca. 
  • Reduced platelet counts were observed in patients.
