Dosage: 

  • For Cholangiocarcinoma, the recommended dosage of Pemazyre is 13.5 mg orally once daily for 14 consecutive days followed by 7 days off therapy, in 21-day cycles. 
  • For Myeloid/lymphoid neoplasm with FGFR1 rearrangement, recommended dosage of Pemazyre is 13.5 mg orally once daily on a continuous basis.

Adverse Reactions:

  • Pemazyre can cause the retinal pigment epithelial detachment which may lead to symptoms such as blurred vision, Photopsia, dry eyes. Dose modification or discontinue the use of pemazyre as per the instructions from the doctor.
  • Pemazyre can cause hyperphosphatemia, which may result in soft tissue mineralization, cutaneous calcification and calcinosis, and non-uremic calciphylaxis. Monitor phosphate levels regularly. If serum phosphate exceeds 5.5 mg/dL, begin a low-phosphate diet. For levels above 7 mg/dL, start phosphate-lowering treatment and consider withholding, reducing the dose, or permanently discontinuing Pemazyre, depending on the severity and duration of the hyperphosphatemia.
